box plot T cell reactivity against SARS-CoV-2 antigens analyzed by FluoroSpot

T cell reactivity against SARS-CoV-2 antigens analyzed by FluoroSpot.

The SARS-COV-2 FluoroSpot results are visualized as Tukey box plots of serologically negative (n = 48) and positive donors (n = 100).

Stimulation indexes (Sls; spot count antigen stimulated sample divided by spot count unstimulated sample) of IFN-y, IL-2 and IFN-y + IL-2 T cell response against SARS-CoV-2 antigens indicate a highly significant difference (**; P value <0.0001) between both groups. P values are calculated by the unpaired, parametric t-test.

The solid line indicates the median, the box indicates the interquartile range (the difference between the 25th and 75th percentiles), the whiskers indicate the range excluding “outliers” and the points indicate outliers, points more than 1.5 IQR from the box.

ROC curve: SARS-CoV-2 IFN-y / IL-2 FluoroSpot

The receiver operating characteristic (ROC) curve shows the true positive rate (sensitivity) versus the false positive rate (100-specificity) of SARS-CoV-2 IFN-γ / IL-2 FluoroSpot for different cut-offs.

Results were compiled by interrogating clinical diagnostics (PCR and serology).

ROC curve: SARS-CoV-2 IFN-y / IL-2 FluoroSpot

T cell reactivity to SARS-CoV-2 vaccines.

SARS-Cov-2 FluoroSpot results (SI; spot count antigen stimulated sample divided by spot count unstimulated sample) before vaccination (TO), two weeks after the first vaccination (T1) and two weeks after the second vaccination (T2) of 11 donors are visualized as box plots.

Data points obtained from each donor are connected by lines.
